Nicole from the Wong Lab at MIT studies how different tissues encode tissue-specific T-cell decisions balancing inflammation and tolerance. Using 25-plex IBEX imaging in lymph nodes draining various organs, she found a hierarchical logic: dendritic cells carry most tissue-specific variation, regulatory T cells next, and T cells least. Principal component analysis suggested distinct mucosal, skin, lung, and gut biases. Interstitial fluid analysis showed cytokine and chemokine patterns matched these regional differences. Treg depletion triggered stronger effector T-cell commitment in mucosal lymph nodes, indicating these sites are primed for inflammation.
