Video Summary
The talk showed that aging reduces Nrf2 in microglia, which shifts them from a homeostatic state to a primed, disease-associated phenotype. This loss of Nrf2 increased inflammatory markers, reduced antioxidant genes, and impaired cognition in mice. Microglia-specific Nrf2 deletion also amplified trained immunity responses to LPS, with higher TNF-α and IL-1β and lower IL-10, indicating stronger immune memory and chronic neuroinflammation. Overall, the study suggests Nrf2 is a key protective regulator during aging and may be a therapeutic target to limit microglial overactivation and age-related cognitive decline.
