The speaker presented a universal CAR T-cell platform called SNAP-CAR that uses adapter antibodies to bind tumor cells indirectly. This design allows better control of activity, targeting of multiple antigens, and potentially reduced toxicity. In preclinical tests, SNAP-CAR T cells showed antigen-specific killing and cytokine production against targets such as CD20, EGFR, and HER2, with strong results in liquid tumors and moderate activity in solid tumors.<br /><br />To improve efficacy, the team repurposed antibody-drug conjugates (ADCs) as adapters by attaching a benzoguanine tag. These BG-ADCs enhanced SNAP-CAR killing in vitro and in vivo, worked against drug-resistant tumor cells, and did not appear toxic to the T cells. They also developed a direct payload-recognition CAR that binds DM1/DM4, which similarly boosted tumor killing and overcame resistance. Overall, the work suggests ADCs can serve as effective CAR T adapters and may improve anti-tumor responses.
