Video Summary
The project investigated how IgE B-cell receptor density affects germinal center survival and differentiation. Using CRISPR, the team deleted IgE’s secreted polyadenylation site, creating “IgE SPAD” mice with higher surface IgE BCR and no secreted IgE. These mice showed fewer IgE germinal center B cells and plasma blasts. Mixed bone marrow chimeras and BCL2 experiments suggested the plasma blast defect is not due to apoptosis alone. In contrast, increased IgE BCR density reduced germinal center apoptosis, confirmed in a separate mouse model. Overall, higher BCR density improves IgE germinal center survival without changing allergic effector responses.
