Researchers developed BEACON, a mucosal adjuvant for genital herpes vaccination, to improve immune responses in the female reproductive tract. By electrostatically combining the TLR9 agonist CpG with chemokines, BEACON targets antigen-presenting cells, reduces neutrophil-driven inflammation, and helps recruit and retain CD8 tissue-resident memory T cells. In mice, vaginal boosting with BEACON plus glycoprotein D generated stronger local antibodies, better long-term survival, and reduced latent infection in dorsal root ganglia compared with intramuscular boosting. The work suggests that local mucosal immunity, especially coordinated B cells and resident CD8 T cells, is key for effective HSV protection.
