The speaker described efforts to develop a pan-arenavirus T cell vaccine. The team mapped human CD4 T cell epitopes across Lassa virus proteins, identified conserved T cell epitope regions (CDERs), and showed these regions are cross-reactive across multiple old-world arenaviruses. LCMV infection in humans generated memory CD4 T cells that recognized Lassa-based CDERs. They then designed AI-based mRNA-LNP vaccines using these CDERs and tested them in mice, where they induced strong, polyfunctional Th1 responses, especially from the L construct. Cross-reactivity to LCMV in mice was limited, guiding further vaccine optimization.
