Pablo Guaman-Tibon described how CCR4 shapes thymocyte development by helping cells interact with dendritic cells in the thymus. Using live imaging and TCR repertoire sequencing, his lab found that CCR4 promotes negative selection of self-reactive thymocytes and helps remove harmful TCRs from the conventional CD4 repertoire. CCR4 also supports Treg generation, especially in models where dendritic cells present antigen. However, CCR4 may also help mature resident Tregs compete with new Tregs for niche access. Overall, CCR4 modulates thymic APC interactions to enforce self-tolerance and influence Treg development, though some knockout effects remain complex and require further study.
