Video Summary
The talk examined ulcerative colitis patients colonized with toxin-negative C. difficile using single-cell RNA sequencing. Even without toxin production, C. difficile-positive mucosa showed increased monocytes, especially CXCL8- and CXCL10-expressing inflammatory monocytes, in both non-inflamed and inflamed tissue. These monocytes displayed activation, antigen presentation, migration, and neutrophil-recruitment programs, while macrophages also showed inflammatory changes. The findings suggest subclinical C. difficile colonization may prime mucosal inflammation in UC and worsen disease. Targeting CXCL8-positive monocyte recruitment could be a therapeutic strategy, though neutrophil validation and larger cohorts are needed.
