Dr. Giuseppe Sangiorgio presented an in vitro platform to model thymic stromal defects using induced pluripotent stem cells (iPSCs) differentiated into mesenchymal stem cells (MSCs). He studied patients with TBX1 mutation, trisomy 21, DiGeorge syndrome, and CHARGE syndrome, generating iPSC-derived MSCs and thymic mesenchymal cells from tissue samples. Transcriptomic analyses showed distinct cell populations and a pro-fibrotic collagen/extracellular matrix program, especially in DiGeorge syndrome. He aims to combine these MSCs with epithelial cells to build isogenic artificial thymic organoids for studying thymus biology and primary immunodeficiencies.
