The talk described how the nuclear receptor NrnB1 (RL-alpha/retinoic acid receptor alpha) regulates CD8 T cells in mice. Nuclear receptors sense lipid-like ligands and can control gene expression through DNA binding, tethering to other proteins, or rapid signaling. NrnB1 expression increases as T cells differentiate and is influenced by local retinoic acid levels, which vary by tissue. Using reporter mice, the speaker showed high retinoic acid in gut and lung, but lower levels in spleen and tumors. <br /><br />Functionally, high NrnB1 suppressed CD8 T-cell infiltration, expansion, and effector programs in tumors and during viral infection. NrnB1 knockout improved anti-tumor immunity and viral control, while transgenic overexpression worsened outcomes. Mechanistically, NrnB1 acted through transactivation and epigenetic control, lowering HAT activity and favoring repression. Overall, retinoic acid reduced NrnB1, and NrnB1 acted as a negative regulator of CD8 T-cell differentiation and function.
