The speaker studied how the E3 ligase Peli-1 affects TLR7-driven lupus-like B-cell responses. In lupus models, loss of Peli-1 increased TLR7 expression, germinal center formation, plasma cells, autoantibodies, ANA positivity, and B-cell metabolism. B-cell-specific knockout showed these effects were intrinsic to B cells, without major T-cell changes. RNA-seq and validation experiments suggested Peli-1 suppresses NFAT5 downstream of TLR7. Overall, Peli-1 acts as a brake on autoimmune B-cell activation and may be a potential therapeutic target in lupus.
