The speaker studied germinal center-independent memory B cells in mice to understand whether they can generate durable antibody responses and protection. Using knockout mouse models, single-cell RNA sequencing, and newly identified surface markers CD148 and CD150, they distinguished germinal center-derived from extrafollicular memory B cells. They found some GC-independent memory cells are T cell-dependent, show low somatic hypermutation, and resemble human extrafollicular B cells. Vaccination and infection experiments showed these cells can mediate antibody-based protection against both matched and mismatched influenza challenges, and transferred memory B cells alone were protective.
