She explained that current immunosuppressants and biologics are often broad, costly, and ineffective for some patients, while T-reg cells offer a more specific way to restore immune balance. GentlyN3.2 is designed around four pillars: stability, growth support, immune evasion, and gut targeting. The platform uses engineering to maintain FOXP3 expression, provide tunable IL-2 signaling, evade T- and NK-cell rejection, and direct cells to the colon and ileum. She described preclinical data showing that the cells remain stable, do not become pro-inflammatory, suppress effector T-cell proliferation, and localize to inflamed gut tissue. In mouse colitis models, the therapy reduced pathology and inflammatory markers. Overall, the talk highlighted GentlyN3.2 as a potentially scalable, long-lasting treatment for Crohn’s disease and ulcerative colitis.
