Hui-Juan from Dartmouth presented NR-Seq, a DNA barcode-based nuclear receptor reporter library designed to overcome limitations of traditional nuclear receptor assays. Nuclear receptors are important transcription factors involved in metabolism, immune regulation, and disease, and many still lack known ligands. Her system uses stable transduction of all-in-one reporter plasmids with dual barcodes, enabling both single-plex flow validation and multiplex bulk RNA-seq readouts. The library showed strong, specific, and stable responses over eight weeks. Future applications include screening bioassays, gut microbial metabolites, drug repurposing, and eventually in vivo reporter models in mice.
