The speaker reviewed how gut immune tolerance was first linked to inflammatory bowel disease through early knockout mouse studies and the microbiota’s role in triggering inflammation. The talk then focused on a newer paradigm: innate lymphoid cells, especially RORγt+ ILC3s that act as antigen-presenting cells in the intestine. These cells use MHC class II to restrain microbiota-specific T cells and promote a specialized IL-10–producing regulatory T-cell population. They also showed that B7 co-stimulation unexpectedly suppresses this regulatory pathway, while CTLA-4 Ig can enhance gut regulatory T cells in certain models. However, this effect depends on the presence of RORγt+ antigen-presenting cells, helping explain why CTLA-4 Ig failed in inflammatory bowel disease trials.
