Tim Springer reviewed the discovery and biology of integrins, beginning with early work on leukocyte adhesion molecules such as LFA-1 and ICAM-1, and how these findings explained T cell contact, leukocyte trafficking, and immune sensitivity. He described how integrins are essential for leukocyte rolling, firm adhesion, and tissue entry, and how defects in these molecules cause immune disease and recurrent infections. Springer then explained integrin structure, conformational states, and how actin-driven force and ligand binding produce ultrasensitive activation. He emphasized that integrins do not simply adhere cells, but also transmit signals that regulate survival, growth, and differentiation.<br /><br />The talk then shifted to organoids. Springer showed that integrin agonists or inhibitory antibodies can strongly alter organoid polarity, survival, and growth. He highlighted bacterial invasin as a powerful integrin-binding tool that can promote organoid formation and may be useful for tissue engineering. Finally, he proposed developing new integrin-specific agonists covalently attached to hydrogels to improve 3D-printed, organ-like tissues for transplantation.
