Majd from Cell Signaling Technology presented IntraSeq, a technology that combines intracellular protein profiling with single-cell RNA-seq using DNA-conjugated antibodies on the 10X Chromium platform. He showed that it can measure surface, intracellular, nuclear, and phospho-proteins alongside transcripts. In CAR-T/Jurkat co-culture experiments with Raji cells, IntraSeq identified activated CAR-T clusters, increased AKT and AP1 signaling at the protein level, and MAP kinase-related RNA changes. It also detected increased apoptosis signatures in Raji cells after 24 hours, demonstrating how IntraSeq reveals coordinated proteomic and transcriptomic responses in single cells.
