Video Summary
Elena Moriarty described research linking sleep fragmentation to atherosclerosis. In ApoE mice on a high-fat diet, interrupted sleep increased aortic plaque formation and made plaques less stable, with more necrotic cores and less collagen. Sleep fragmentation also activated neutrophils, boosting ROS, degranulation, and NETosis, and promoted their recruitment to the aorta. Neutrophil depletion reduced plaque burden, but a more targeted STAT4 deletion lowered lesion formation and neutrophil activation while unexpectedly increasing mortality, possibly due to intestinal oxidative stress, leaky gut, and circulating LPS. The team is now investigating gut infection and barrier dysfunction as possible causes.
