Novel Mouse Models Allow Selective Labelling and Depletion of Perinatal and Adult-derived Regulatory T Cells

Sookyong Joo presented new mouse models to selectively label and deplete perinatal versus adult-derived regulatory T cells (Tregs). Existing tamoxifen-based methods have drawbacks, especially in newborns and because they label all Tregs present during treatment. Her lab created two complementary models: an “early” model that labels Tregs before tamoxifen exposure and an “late” model that labels Tregs arising after exposure. She validated both models in normal mice and bone marrow chimeras, showing efficient and specific GFP labeling. She also demonstrated that the early model can timestamp other perinatally derived immune cells, including small intestine CD8αα T cells and B1a B cells. Using the early model, she found perinatally derived Tregs persist in many tissues into adulthood, and their frequency remains fairly stable from 16 to 24 weeks, though blood Tregs shift over time toward adult-derived cells. Finally, diphtheria toxin efficiently depleted labeled Tregs.
