Elise Nguyen presented research on improving flu vaccines by adding TLR4 and TLR9 agonists to a subunit vaccine. Her lab developed IVAX-1, combining CPG, MPLA, and the oil-in-water emulsion Atavax. In mouse H5N1 studies, IVAX-1 rapidly induced strong interferon-gamma-producing CD4 and CD8 T-cell responses, with signs of clonal expansion in lymph nodes. A single dose also protected mice from lethal influenza challenge, reduced weight loss, and lowered lung viral titers by 44,000-fold compared with controls. She concluded that this adjuvant combination strengthens cell-mediated immunity and improves viral clearance, with manageable short-term inflammation.
