The speaker presented a PhD method for cheaply and quickly synthesizing and transducing pooled T cell receptor (TCR) libraries. The goal is to streamline functional validation of TCR hits from single-cell and tetramer-based antigen discovery, since many annotated TCRs are not truly antigen-specific. Their approach, called CREP, uses codon-optimized TRV genes and pooled CDR3 oligos assembled by PCR in plates, allowing specific pairing and reducing cost and time. In early tests, the method showed no dropouts in a 13-TCR library and later worked on a 123-TCR library at about $10 per TCR. Functional screening identified candidate epitope-specific TCRs, and the workflow can be done in a day with standard cloning tools. Future work aims to scale this to cohort-level antigen screening and broader repertoire studies.
