Erin Maul described developing IL-2-conjugated lipid nanoparticles (LNPs) to deliver mRNA specifically to regulatory T cells (Tregs) for autoimmune therapy, especially type 1 diabetes. She compared IL-2 targeting with anti-CD25 antibody targeting and found IL-2 LNPs were more effective and selective in mice and human cells. The system showed strong Treg uptake, limited off-target delivery, reduced liver accumulation, and signs of IL-2 receptor signaling. This approach could avoid laborious ex vivo Treg engineering and may be adapted to deliver cargos that improve Treg function and pancreatic homing.
