Dr. Jacqueline Nestor described building a 40-million-cell blood immune atlas from over 5,000 samples across 40 disease conditions, including autoimmune, infectious, and checkpoint inhibitor toxicity cases. Using single-cell RNA, protein, and TCR data, her team optimized integration and identified shared and disease-specific immune cell states. Preliminary findings showed known biology, like recent thymic immigrant CD4 T cells being more common in children, and revealed cytotoxic T cell subsets enriched in diseases such as lupus, diabetes, and celiac disease. Autoimmune risk variants were also most active in these cytotoxic cells, suggesting common mechanisms across autoimmune disorders.
