He focuses on naïve B cells and the expanded double-negative 2 (DN2) population, which is linked to autoreactive, antibody-secreting cells. His lab found that TLR7 promotes DN2 and plasmablast expansion, while TLR9 can expand DN2 cells even more strongly in some cultures and may restrain plasmablast/plasma cell differentiation in tonsil organoids. Using flow cytometry and single-cell RNA sequencing, the team hopes to define these pathways and assess TLR9 as a potential therapeutic target in SLE.
