The speaker presents SB2H, a highly selective STING degrader designed to eliminate STING rather than merely inhibit it. STING drives inflammation in interferonopathies, autoimmune disease, and some metabolic disorders, but existing inhibitors are weak or nonselective. SB2H rapidly and potently depletes STING in human cells, patient fibroblasts, and PBMCs, with minimal off-target effects and good safety. In mice, it shows favorable pharmacokinetics, suppresses STING-driven cytokines, reduces inflammation, improves survival in Trex1 knockout mice, and improves disease in a SAVI model. The compound is now being evaluated for clinical development in STING-related inflammatory and fibrotic diseases.
