Taishi Kimura of HDT Bio described Lion, a cationic RNA delivery system that binds RNA on its surface and appears better tolerated than lipid nanoparticles. Using localized RepRNA vaccination in mice, the team found that muscle becomes an active immune niche, recruiting CXCR5+ CD4 T cells and B cells. CD4 T cells were required for antibody responses, and plasmablasts expanded mainly in muscle. Early responses depended partly on IL-21 and germinal centers, but later antibody production persisted without them. Blocking lymphocyte trafficking had little effect, showing localized intramuscular RepRNA can drive antibody immunity independently of continuous lymph node recirculation.
