The talk introduced SlideGoTags, a spatial genomics method that combines SlideTags with TCR sequencing and targeted neoantigen genotyping to link tumor antigens with cognate T cell clonotypes in tissue. Using mouse MC38 tumors expressing known neoantigens and OT1 T cells, the method detected co-localization between mutant tumor cells and antigen-specific TCRs, and showed that anti-PD-1 treatment promoted a proliferative neoantigen-specific T cell niche. In a human clear cell renal cell carcinoma sample, SlideGoTags mapped neoantigen-specific and tumor-specific TCRs alongside mutant tumor cells, revealing an immunogenic niche enriched for strong-avidity TCRs, exhausted T cells, interferon signaling, and antigen presentation genes. Across additional human tumors, similar interferon-rich niches varied by cancer type, with RCC and melanoma generally higher than GBM and ovarian cancer. Overall, the study shows that spatial proximity can help infer TCR-neoantigen specificity and reveals how immune niches shape anti-tumor responses.
