Video Summary
Cheng, a Cambridge PhD candidate, presented research on human lymph node metastasis in triple-negative breast cancer. Using sentinel and secondary lymph nodes from 15 treatment-naive patients, the team built a single-cell and spatial immune atlas from CD45+ cells. They found progressive loss of dendritic cells, especially CD1C+ cDC2s, and NK cells, alongside expansion of immunosuppressive macrophages. CD1C+ cDC2 abundance in lymph nodes predicted better response and longer event-free survival in anti-PD1-treated patients. Machine-learning analyses and mouse studies suggested that combining anti-MARCO with PD-1 blockade may improve tumor control.
