The speaker described research showing that TRIM21 promotes inflammasome activation by binding ASC, ubiquitinating it, and enabling ASC speck formation, polymerization, and caspase-1 activation. TRIM21 knockout cells had reduced IL-1β, IL-18, gasdermin cleavage, and pyroptotic death, while in mice TRIM21 deficiency rescued autoinflammatory disease phenotypes. TRIM21 was also found in released ASC specks and elevated in CAPS and FMF patient samples, along with anti-TRIM21 autoantibodies. Overall, TRIM21 appears to drive autoinflammatory pathology and may link autoimmune and autoinflammatory disease mechanisms.
