The talk described how mast cell progenitors are recruited into inflamed lungs during type 2 inflammatory disease. Using a house dust mite mouse model and human/mouse single-cell data, the speaker identified a conserved EBI2–CH25H axis: endothelial CH25H produces the EBI2 ligand, which attracts progenitor cells expressing EBI2. In vitro migration was blocked by an EBI2 inhibitor. In vivo, deleting CH25H or EBI2 prevented mast cell progenitor recruitment and expansion in lung tissue. The findings suggest mast cell maturation begins earlier than expected and may involve a vascular pre-maturation stage before tissue entry.
