Jeff Yoder from North Carolina State University presented research on the diversification of CD300 immune receptor genes across mammalian species. His lab studies how immune systems distinguish self from non-self, focusing on CD300 receptors, which can be activating or inhibitory and often bind phospholipids such as phosphatidylserine. Using genome databases, the team found strong conserved synteny for the CD300 gene cluster across vertebrates, but large differences in gene number among species. Marine mammals, especially cetaceans, show marked CD300 loss compared with terrestrial relatives like camels, while cetaceans also appear enriched for inhibitory receptors. <br /><br />Yoder’s group developed a new lipid-binding assay using recombinant CD300 fusion proteins and mass spectrometry to identify bound lipids from natural extracts. Early results suggest substantial differences between human CD300A and CD300F, while some nonhuman orthologs resemble CD300A. The work aims to link molecular evolution with changes in lipid-binding function and to reconstruct ancestral CD300 proteins for future testing.
