Video Summary
Shachi Vyas described research on ARID5A, an RNA-binding protein induced during CD4 T-cell activation. While ARID5A is known to stabilize inflammatory mRNAs, her lab found it also promotes mRNA translation. ARID5A knockout T cells showed reduced protein synthesis and impaired proliferation, but normal activation markers, signaling, and apoptosis. Ribosome profiling revealed decreased translation of ribosomal and initiation-factor mRNAs. Polysome profiling and co-immunoprecipitation suggested ARID5A associates with EIF4G and the 40S ribosomal subunit to support translation initiation. The work links ARID5A to T-cell expansion and may inform therapies for autoimmune inflammation.
