Dr. James Allison recounts his Texas roots and early inspiration for science, shaped by excellent teachers and the death of his mother from lymphoma, which motivated his desire for better cancer treatments. After entering the University of Texas at Austin at 16 intending to pursue medicine, he became drawn to research through lab work, gravitating toward immunology and cancer models. Draft pressure during graduate school influenced him to work on the leukemia drug asparaginase, and after earning his PhD he pursued postdoctoral work at Scripps, studying MHC molecules as immunology pivoted toward understanding T cells.<br /><br />Back in Texas at a UT/MD Anderson facility near Smithville, Allison helped identify the T-cell antigen receptor using biochemical and antibody-based approaches—an initially controversial finding later confirmed by other methods. Recognizing the need for molecular biology resources, he spent time at Stanford and then moved to UC Berkeley, where he built immunology within Molecular and Cell Biology and expanded shared research infrastructure.<br /><br />At Berkeley his lab demonstrated CD28’s role in T-cell costimulation and, crucially, argued that CTLA-4 is an inhibitory “brake,” not a stimulator—a view confirmed by knockout mice. This led to the idea of blocking CTLA-4 to unleash anti-tumor immunity; mouse studies showed dramatic tumor regression and lasting immunity. To translate the work clinically, he moved to Memorial Sloan Kettering, supporting development and mechanistic study of ipilimumab (anti–CTLA-4), which produced durable survival “tails” in metastatic melanoma.<br /><br />Allison later returned to MD Anderson to build an “immunotherapy platform” to accelerate rational combination trials and immune monitoring. He also reflects on AAI leadership, funding challenges, mentorship, and his lifelong connection to music, including playing harmonica in a scientist band called “The Checkpoints.”

Smith Distinguished Chair of the Department of Immunology, Co-Director of the Parker Institute for Cancer Immunotherapy, and Executive Director of the Immunotherapy Platform at The University of Texas MD Anderson Cancer Center. He is also Deputy Director of the Koch Center for Applied Research of Genitourinary Cancers at MD Anderson. Dr. Allison was President of The American Association of Immunologists from 2001 to 2002 and served as an AAI Council member from 1996 to 2003. He was awarded the AAI-Dana Foundation Award in Human Immunology Research in 2008 and the AAI Lifetime Achievement Award in 2011, and elected a Distinguished Fellow of AAI in 2019.
